چکیده
UNLABELLED PREMISE OF THE STUDY Focused ion beam-scanning electron microscopy (FIB-SEM) combines the ability to sequentially mill the sample surface and obtain SEM images that can be used to create 3D renderings with micron-level resolution. We have applied FIB-SEM to study Arabidopsis cell architecture. The goal was to determine the efficacy of this technique in plant tissue and cellular studies and to demonstrate its usefulness in studying cell and organelle architecture and distribution. • METHODS Seed aleurone, leaf mesophyll, stem cortex, root cortex, and petal lamina from Arabidopsis were fixed and embedded for electron microscopy using protocols developed for animal tissues and modified for use with plant cells. Each sample was sectioned using the FIB and imaged with SEM. These serial images were assembled to produce 3D renderings of each cell type. • RESULTS Organelles such as nuclei and chloroplasts were easily identifiable, and other structures such as endoplasmic reticula, lipid bodies, and starch grains were distinguishable in each tissue. • DISCUSSION The application of FIB-SEM produced 3D renderings of five plant cell types and offered unique views of their shapes and internal content. These results demonstrate the usefulness of FIB-SEM for organelle distribution and cell architecture studies.

A method for preparing spaceflight RNAlater-fixed Arabidopsis thaliana (Brassicaceae) tissue for scanning electron microscopy1
UNLABELLED PREMISE OF THE STUDY In spaceflight experiments, tissues for morphologic study are fixed in 3% glutaraldehyde, while tissues for molecular study are fixed in RNAlater; thus, an experiment containing both study components requires multiple fixation strategies. The possibility of using RNAlater-fixed materials for standard SEM-based morphometric investigation was explored to expand ...
متن کاملGene transcriptomic profile in arabidopsis thaliana mediated by radiation-induced bystander effects
Background: The in vivo radiation-induced bystander effects (RIBE) at the developmental, genetic, and epigenetic levels have been well demonstrated using model plant Arabidopsis thaliana (A. thaliana). However, the mechanisms underlying RIBE in plants are not clear, especially lacking a comprehensive knowledge about the genes and biological pathways involved in the RIBE in plants. Materials and...
متن کاملThree-Dimensional Architecture and Biogenesis of Membrane Structures Associated with Plant Virus Replication
Positive-sense (+) RNA viruses represent the most abundant group of viruses and are dependent on the host cell machinery to replicate. One remarkable feature that occurs after (+) RNA virus entry into cells is the remodeling of host endomembranes, leading to the formation of viral replication factories. Recently, rapid progress in three-dimensional (3D) imaging technologies, such as electron to...
متن کاملHuman cardiac telocytes: 3D imaging by FIB-SEM tomography
Telocyte (TC) is a newly identified type of cell in the cardiac interstitium (www.telocytes.com). TCs are described by classical transmission electron microscopy as cells with very thin and long telopodes (Tps; cellular prolongations) having podoms (dilations) and podomers (very thin segments). TCs' three-dimensional (3D) morphology is still unknown. Cardiac TCs seem to be particularly involved...
متن کاملNovel characteristics of normal supraspinatus insertion in rats: an ultrastructural analysis using three-dimensional reconstruction using focused ion beam/scanning electron microscope tomography.
BACKGROUND the histological architecture of the insertion after a rotator cuff repair is completely different from that of normal tendon-bone insertions. Analysis of normal insertions by electron microscopy may enhance the understanding of the pathophysiology of tendon-to-bone healing after rotator cuff repair. The present study examined the normal supraspinatus insertion in rats using a new th...
